What is an RNA-LNP ?

RNA-LNPs (RNA–lipid nanoparticles) are nanoscale delivery systems composed of four key lipid components: ionizable lipids, cholesterol (sterol), phospholipids, and PEG-lipids. They are specifically engineered to encapsulate and deliver nucleic acids such as mRNA or siRNA.

RNA-LNPs are considered the gold standard for nucleic acid delivery because they:

  • Protect sensitive RNA cargo from enzymatic degradation
  • Facilitate efficient cellular uptake and endosomal escape
  • Can be tailored in size, charge, and composition to optimize efficacy, safety, and biodistribution

What can be encapsulated into LNP?

LNPs can encapsulate hydrophilic, charged, or fragile biomolecules or nucleic acids, including:

Optimizing the biodistribution and tissue specifity of RNA-LNPs can be achieved through two main strategies:

1. Passive targeting
By tuning intrinsic LNP properties—such as particle size, lipid composition, and lipid ratios—the natural distribution of RNA-LNPs in the body can be influenced.
It’s the most straightforward approach but requires broad screening of lipid formulations to identify optimal profiles

2. Active targeting
By decorating LNPs with targeting ligands (e.g., antibodies, peptides, aptamers), delivery can be directed toward specific tissues or cell types.
It’s an overall more complex design and validation process but enables a more precise, tissue-specific delivery.
