What is an RNA-LNP ?

RNA–lipid nanoparticles (RNA-LNPs) are nanoscale carriers built from four main lipid components: ionizable lipids, cholesterol, phospholipids, and PEGylated lipids. Their design enables the encapsulation and delivery of nucleic acids such as mRNA or siRNA.

Recognized as the current benchmark in nucleic acid delivery, RNA-LNPs offer several advantages:

  • Maximize RNA encapsulation
  • Shield fragile RNA molecules from enzymatic breakdown
  • Promote efficient cellular internalization and release from endosomes
  • Allow fine-tuning of particle size, charge, and composition to balance safety, efficacy, and biodistribution

What can be encapsulated into LNP?

Lipid nanoparticles (LNPs) are versatile carriers capable of encapsulating a wide range of hydrophilic, charged, or delicate biomolecules and nucleic acids, such as:

  • mRNA, saRNA, CircRNA – used in vaccines and protein replacement therapies
  • CRISPR-Cas components– including RNP complexes or mRNA combined with gRNA
  • siRNA, miRNA, ASO  – for gene silencing applications in oncology and rare diseases

Does the training cover targeting strategies for RNA-LNPs?

During the training, we provide an introduction to strategies for optimizing RNA-LNP specificity and biodistribution. Two main approaches are discussed:

  1. Active targeting – In more advanced designs, LNPs can be functionalized with targeting moieties (e.g., antibodies, peptides, or aptamers) to guide delivery toward specific tissues or cell types. Although this strategy requires additional design, characterization, and validation steps, it enables greater precision and tissue selectivity.
  2. Passive targeting – By tuning intrinsic LNP parameters such as particle size, lipid composition, and lipid ratios, it is possible to influence how nanoparticles naturally distribute within the body. This approach is relatively straightforward and widely used, though it generally relies on extensive screening to identify the most effective formulation profiles.
